Good morning,

is it possible to make a configuration in which the DHCP server only
responds when there's no option125 provided within the discover/request/..?

Yes,

use dhcp-match to set a tag when option 125 is present, and dhcp-ignore
to ignore requests that don't set that tag.

dhcp-match=set:haveoption,125
dhcp-ignore=tag:!haveoption
